HuProt?人全蛋白質(zhì)組芯片
HuProt? HuProtTM V4.0芯片上有超過2萬個釀酒酵母表達(dá), 帶有GST和His標(biāo)簽的重組人蛋白和部分小鼠重要蛋白, 其中89%為全長蛋白,涉及約17000個人類基因和124種鼠對照蛋白。酵母蛋白具有翻譯后修飾, 故也適用于PTM研究。非接觸噴印 (Arrayjet Non-Contact Piezoelectric Print) 保證了樣品點(diǎn)均勻度和復(fù)點(diǎn)間一致性, 分析結(jié)果更準(zhǔn)確。
檢測時,分析物(Analyte)先與芯片孵育, 并與特定蛋白結(jié)合。分析物是血液IgG或雜交瘤培養(yǎng)上清時, 使用熒光標(biāo)記的抗人(鼠)IgG抗體即可檢出結(jié)合的IgG, 對其他分子則通過標(biāo)記分析物或其抗體的方式檢測,熒光掃描芯片取得數(shù)據(jù)后用于分析, 后續(xù)針對初篩得到的蛋白定制小規(guī)模芯片進(jìn)行大樣本驗(yàn)證。
